Effect of WuWeiShiGaoTang on OVA induced allergic rhinitis in Balb/c

Objective: To explore the protect effect of Wuwei Shigao Tang on Allergic rhinitis (AR) mice induced by ovalbumin (OVA)
Methods: 40 BALB/c female mice were randomly divided into 5 groups:blank control group. OVA model group. positive drug group (loratadine 10 mg/kg). and Wuwei Shigao Tang low dose group (5.46 g/kg). High-dose group (10.92 g/kg). 8 mice for each group. Except for blank control group. OVA was used to prepare AR model. The nasal symptoms were evaluated by observing the scratching nose and sneezing in mice. Serum immunoglobulin E (IgE). tumor necrosis factor (TNF) α. interleukin 1β interleukin-4 and interferon γ were detected by ELISA. Western blot was used to detect the expression of T-bet and GATA-3 in nasal mucosa.
Results: Compared with the blank control group. the allergic rhinitis symptoms of AR model mice were obvious.the infiltration of eosinophils was significant, and the level of IgE. TNF-α. IL-1β and IL-4 in serum were significantly increased. while the level of IFN-γ decreased significantly. T-bet protein in nasal mucosa significantly decreased (P < 0.01)and GATA-3 expression increased (P < 0.001). However. Wuwei Shigao Tang could significantly improve nasal allergic symptoms in mice (P < 0.01). decrease serum IgE TNF-α IL-1β and IL-4 levels.Increased serum IFN-γ levels. increase the expression of T-bet in nasal mucosa and decrease the expression of GATA-3 (P < 0.01).
Conclusions: Wuwei Shigao Tang has the therapeutic effect on OVA-induced AR in mice. which is equivalent to the treatment of positive drug. The mechanism may be related to the regulation of Th1/Th2 immune imbalance.
