A New Phase of Organoid Culture: EMT, CSC, Exosome, and HSP

Two-dimensional (2D) cell culture system by using plastic wares made innovation and has been often used in biological research, including drug screening; however, the 2D culture system forces malignant transformation and integrin expression on cancer cells. In contrast, cells can form three-dimensional (3D) aggregates, organoids or tumoroids in 3D culture environments. We have examined alteration of cellular and organoid morphologies, intra-tumoral hypoxia levels, epithelial-mesenchymal transition (EMT), gene expression profiles and secretion of extracellular vesicles (EVs) or exosomes by culturing cancer cells in 2D and 3D matrix-free NanoEnvironments (1, 2). One of our studies clarified a novel EMT inhibitor through a novel drug screening system using NanoCulture Plates (1). Our recent study further clarified a novel aspect of tumoroids with properties of cancer stem-like cells (CSCs) that secrete particular EVs and heat shock proteins (2).
