Beneficial effects of Ginsenosides Rg3 on cyclophosphamide-induced immunosuppression in mice

Objective: Side effects caused by chemotherapy usually lead to decline in the quality of life for the patients and even have to discontinued because of intolerance. The immunosuppression caused by chemotherapy has been paid more attention in recent years, ginsenosides Rg3 (Rg3) have been suggested to contribute to enhance immune function.
Method: In this study, we established an immunosuppression BABL/c mice model with cyclophosphamide (CTX). The mice in experimental groups were orally ingested once daily with two doses (7.5mg/kg and 15mg/kg) of Rg3 for 19 consecutive days, respectively, accompanied by intraperitoneal injection of CTX (80mg/kg) on the 15th-19th days. CTX group i.g. saline from 1st-14th days and i.p. CTX (80mg/kg) on the 15th-19th days. Saline was used as control.
Results: Compared with the CTX group, the two experimental groups could recovery weight, spleen and thymus indices, the counts of WBC, RBC, LY%, PLT in peripheral blood, increased the activities of LDH and ACP in spleen tissues. Furthermore, the experiment groups restored the level of interleukin-2 (IL-2), immunoglobulin G (IgG), granulocyte colony-stimulating factor (G-CSF) in the serum. Pathology showed that the lymphoid follicles and germinal centers in the spleen of the experiment groups were increased significantly, the number of lymphocytes in the lobuli thymi increased and the arrangement were tight. The results of immunohistochemistry showed that the expression of CD3+, CD4+ in spleen and thymus cell surface were increased significantly, CD8+ was decreased significantly. The expression of ICOS mRNA in spleen and thymus cells from experiment groups was up-regulated compared with model group, the expression of PD-1 mRNA was decreased, however, the expression of OX40 mRNA doesn't change in the experiment groups.
Conclusion: The result demonstrated Rg3 could recovery of immunosuppression in CTX-treated mice, in particular, it has a certain therapeutic value to reduce the side effects in the treatment of tumor.
