XANTHINE OXIDASE INHIBITOR EFFECT OF 3-ETHYL-4HYDROXY- 5 METHYLBENZALDEHYDE ISOLATED FROM ROSELLE CALYX (HIBISCUS SABDARIFFA L.)

Objective: The objective of the research determines to isolate the active compound from ethyl acetate fraction of roselle calyx and its activity as xanthine oxidase inhibitor.

Methods: Isolation of active compound from the ethyl acetate fraction of roselle calyx was using Sephadex LH-20, and the activity of xanthine oxidase inhibitory effect was determined by measuring uric acid formation in UV Spectrophotometry

Result: Isolation of active compound from ethyl acetate fraction of roselle calyx using Sephadex LH- 20 produced isolates that have a role in anti-hyperuricemia activity. Identification of isolate was based on the UV, NMR, FTIR Spectrophotometry analysis and showed that the isolate was 3 -ethyl -4 hydroxy-5-methyl benzaldehyde. In vitro assay demonstrated that the isolate inhibited xanthine oxidase and its ability as xanthine oxidase inhibitor was relatively good compared to the fraction which isolates 100 µg/ml at 34.07 % respectively, with IC50 isolate was 143.45 µg/ml.

Conclusion: 3-ethyl-4 hydroxy-5methylbenzaldehyde isolated from roselle calyx has xanthine oxidase inhibitor activity.
